Alkyne peg NHS (alkynyl polyethylene glycol active ester) is a bifunctional peg derivative, which can be used to modify proteins, peptides and other substances through dynamic combinatorial chemistry. Alkynyl groups in alkyne peg NHS can react with azides in aqueous solution catalyzed by copper. Through stable amide bond formation, alkyne peg NHS can simply and effectively bind terminal alkyne groups to primary amine groups (such as lysine residues or side chains of aminosilane coated surfaces).
Hydrophilic short polyethylene glycol (PEG) spacer arms enhance the water solubility of labeled molecules and reduce the aggregation of labeled proteins stored in solution. In addition, it provides a long and flexible spacer, which can minimize the steric hindrance in the connection with azide counterparts.
